There are two ways to buy them: you can buy them on the spot (having to stand in a very long waiting line), or pre-purchase them on the internet on the official website of the Vatican Museums.
The museum’s website provides visitors with the official ticket presale service which will allow you to access without having to queue at the entrance. However, online tickets will be vouchers that you must show to the service personnel outside the Museums, pass the security check and then go to the reserved cash desks or use the electronic ticket machines, which will print your ticket with which you can go to the turnstile.
Unfortunately you have to organize your visit well in advance if you want to buy online, because the tickets available are limited! In fact, it is not said that you will always be able to find them, you can still buy entry on other sites that have a wide availability of “skip the line” tickets, which however have a higher price than those sold on the official site.
